The increasing development of the biotechnologies, and his application on numerous plants and other organism have aroused in the last years a series of worries and question on the potential risks connected with the introduction of such organism in the environment. In this context the project has been addressed to the effect of the GMO on the difference of soil microrganism. A perturbation of natural condition due to GMO can insert stress factor that influence the structure and the microbial activity of soil, with consequences and dangerous effects in the middle and long period.

Il crescente sviluppo delle biotecnologie e della sua applicazione su numerose varietà vegetali ed altri organismi, ha suscitato negli ultimi anni una serie di preoccupazioni ed interrogativi sui potenziali rischi connessi con l'introduzione di questi organismi nell'ambiente. In questo contesto, il progetto è stato indirizzato all'effetto delle OGM sulla diversità dei microrganismi del suolo. Una perturbazione delle condizioni naturali dovuta agli OGM può indurre a fattori di stress che influiscono sulla struttura e sull'attività dei microrganismi del suolo, con conseguenze nel medio e lungo periodo.

The main aim of this work was to study the effect of different organic amendments in the rhizosphere soil with innovative techniques, and the building of a modelling tool to describe the carbon flow in the rhizosphere soil. For this reason maize plants were grown in rhizobox systems amended with different organic materials. The available fraction of organic C in each layer was assessed with the lux-marked p. fluorescens 10586 pucd607. Total organic carbon (TOC), organic acid distribution in the rhizosphere and total number of culturable cells as colony forming units (CFU) were also determined. A modelling tool is presented that describes the carbon flow in the rhizosphere. The model solves with the Crank-Nicolson discretisation scheme a diffusion-degradation equation and is coupled to the inverse modelling software pest. Finally a five-compartment conceptual model is proposed for describing organic compounds bioavailabililty in soil.

L'obiettivo di questa tesi di dottorato era lo studio della influenza di diversi amendanti organici sulla rizosfera e la costruzione di un modello matematico per descrivere il flusso di carbonio nella rizosfera. Per questo motivo, plantule di mais sono state seminate e cresciute all'interno di sistemi rhizobox. La frazione disponibile del carbonio in ogni strato è stata valutata con il biosensore luminescente P. Fluorescence 10586 PUCD607. Inoltre sono stati determinati il carbonio organico totale (TOC), la distribuzione degli acidi organici nella rizosfera e la carica microbica totale. Un modello matematico viene presentato che descrive il flusso del carbonio nella rizosfera. Il modello risolve un'equazione di diffusione-degradazione con lo schema di discretizzazione di Crank-Nikolson ed è accoppiato all'inverse modelling software pest. Infine viene proposto un modello concettuale per la descrizione della biodisponibilità delle sostanze nel suolo.

The behaviour evaluation of a pesticide metabolite was divided in two different but complementary studies, using the radiolabeled metabolite: 1. evaluation of aerobic degradation of 14C-metabolite in soil; 2. evaluation of sorption of 14C-metabolite in soil. The study of GLP (Good Laboratory Practices) for multisite studies lead to the management of validation study for determination of glyphosate residues in fruits and vegetables.

La valutazione del comportamento ambientale del metabolita di un pesticida è stato suddiviso in due studi indipendenti ma complementari tra loro utilizzando il metabolita radiomarcato con 14C: 1. Valutazione della degradazione aerobica del 14C-metabolita nel suolo; 2. Valutazione dell'assorbimento e desadsorbimento del 14C-metabolita nel suolo. L'attività di studio delle normative e dei regolamenti concernenti le Buone Pratiche di Laboratorio nell'ambito di studi multisito ed è poi sfociato nella gestione di uno studio di validazione di un metodo di analisi per la determinazione dei residui di glyphosate in frutta ed ortaggi.

The main aim of the research was to monitor the concentrations of nitrates, the herbicide terbuthylazine and its products of the environmental degradation, desethyl-terbuthylazine, in shallow groundwater, according to the normal agricultural practices in different Italian areas representative of maize crop.
The first step was to identify the monitoring sites, therefore it was necessary to create a general procedure to individuate maize representative areas in Italy. Inside these area 11 monitoring sites were selected: 4 in Lombardy, 2 in Emilia Romagna, 2 in Veneto, 2 in Piedmont and one in Friuli-Venezia Giulia.
In April 2005 the monitoring started. Groundwater samplings were carried out every two months. The IDPR index, a tool for national and regional vulnerability mapping, was applied. The index reflects the influence of the subsurface geological formations on the surface water runoff or infiltration.
The distribution probability analysis of the results showed that there was a very low probability of exceeding the limit fixed by law, this probability was higher for the metabolite. The greater probability of exceed the 0.1 μg/L occurred in two sites, both irrigated with basin irrigation system. Nitrate concentrations resulted to be very low in shallow groundwater.
Moreover, it was used a mathematical model which calculate the leaching of pesticides (MACRO 5.1).

Lo scopo principale di questa ricerca è stato di monitorare la concentrazione di nitrati, dell'erbicida terbutilazina e di un suo metabolita, la desethylterbuthylazina, in acque sotterranee superficiali, secondo le normali pratiche agronomiche in differenti aree italiane rappresentative della coltura maidicola.
Il primo passo è stato identificare i siti di monitoraggio, quindi è stata creata una procedura generale al fine di individuare aree rappresentative per il mais in Italia. Dentro tali aree sono stati selezionati dei siti di: 4 in Lombardia, 2 in Emilia Romagna, 2 in Veneto, 2 in Piemonte e uno in Friuli-Venezia Giulia.
In Aprile 2005 il monitoraggio ha avuto inizio e i campionamenti sono stati eseguiti ogni due mesi.
L'indice “IDPR”, strumento per la mappatura della vulnerabilità a livello regionale e nazionale, è stato applicato alle aree oggetto di studio. L'indice riflette l'influenza che le formazioni geologiche del sottosuolo hanno sullo scorrimento superficiale e sull'infiltrazione.
L'analisi della probabilità di distribuzione dei risultati ha mostrato che una bassa probabilità di superare i limiti fissati per legge, questa probabilità è risultata più alta per il metabolita.
Le concentrazioni dei nitrati nelle acque sotterranee sono risultate molto basse.
Inoltre, è stato applicato un modello matematico (MACRO vs 5.1) di previsione che calcola la percolazione dei pesticidi.

Food traceability systems have a broader scope and aim to document the history of a product along the entire production chain from primary raw materials to the final consumable product.
the present work has looked at ways of detecting rare earth and strontium isotope abundance ratios. It is fundamental to have knowledge of the REE concentrations of the soil where the plants are grown and the distribution of the REEs in the different parts of the plants: roots, stems, leaves, and berries.
Tomato is one of the most widely grown vegetables in the world. The analytical procedure, using inductively coupled mass spectrometry (ICP-MS) and HR-ICP-MS
proved to be suitable for the determination of REEs in plants and soil, and was validated using certified samples.
Strontium isotope ratio is an important parameter allowing the discrimination of foods coming from different geographical origins. Very high precision is usually reached with Thermal Ionization Mass Spectrometer (TIMS).
The analytical techniques have permitted to determine all REEs elements in compartments of tomato plants.
The described method point out the measurement of the 87Sr/86Sr ratio as a very important parameter for the discrimination of tomato from different geographical origins, even if the present experimental work treating a limited number of samples have to be considered as a “first step”.

METODI CHIMICI PER IL RICONOSCIMENTO DELL'ORIGINE GEOGRAFICA DEGLI ALIMENTI

Il presente studio affronta il tema della rintracciabilità alimentare di prodotti agroalimentari, analizzando il caso del pomodoro da industria, cercando di trovare degli indicatori chimici (markers) che siano in grado di discriminare prodotti provenienti da aree geografiche differenti. Viene quindi affrontato il tema della tipicità che consiste nel legame esistente tra l'alimento e il territorio nel quale viene prodotto. I parametri chimici che vengono indagati sono elementi appartenenti al gruppo delle terre rare (o serie dei lantanidi) e dei rapporti isotopici degli elementi pesanti quali lo Stronzio. La determinazione analitica delle terre rare nel pomodoro è stata effettuata con ICP-MS e HR-ICP-MS (Inductivity Coupled Plasma Mass Spectrometry, Hight Resolution Inductivity Coupled Plasma Mass Spectrometry). I rapporti isotopici dello Stronzio sono stati determinati con la tecnica TIMS (Thermal-Ionization Mass Spectroscopy)
Le tecniche analitiche utilizzate hanno permesso di determinare con precisione e accuratezza tutti gli elementi appartenenti al gruppo delle terre rare nei comparti della pianta di pomodoro e nel terreno. La misura dei rapporti isotopici dello stronzio ha permesso di discriminare prodotti provenienti da aree geografiche differenti.

Chromium in the trivalent form (CrIII), is an important component of health human and animal, in contrast CrVI is highly toxic and cancinogen and may cause death to animals and humans if ingested in large doses. In the last few years were defined the acceptable limit of chromium content in biosolid or tannery sludge for their use in agricolture. In this work has been studied the possible reutilizable the sludge in agricolture and other residual product of tanning cicle with test in field on wheat and maize. Was used for this purpose fertilizer NP product by cuticles integrate and tanning land plaster (GDC) with test in field on tomato. The object of this work could be evaluation of suitability of tannery land plaster as amenments , adsortion of chromium by plants of tomato and valutation possible ossidation to CrVI from CrIII linked to GDC.

Il cromo, nella forma trivalente (CrIII)è un importante componente per la salute umana e animale, mentre il CrVI è altamente tossico e cancerogeno causando la morte se ingerito ad elevate dosi. Negli ultimi anni sono stati definiti i limiti accettabili del contenuto di cromo nei biosolidi o fanghi di conceria per una loro utilizzazione nel suolo in agricoltura. In questa ricerca si è studiato la possibilità di riutilizzo in agricoltura dei fanghi e di altri sottoprodotti del ciclo conciario con prove in campo su frumento e mais, usando un concime NP prodotto a partire da pellicino integrato. Con prove in campo su pomodoro si è utilizzato il gesso di defecazione (GDC) con l'obbiettivo generale di valutare l'idoneità del gesso ad un impiego ammendante, valutare l'assorbimento di cromo in piante di pomodoro cresciute in suoli trattati con GDC e valutare i possibili fenomeni di ossidazione a CrVI da CrIII legato al GDC.
